Multiplicity for a group sequential trial with biomarker subpopulations

چکیده
Biomarker subpopulations have become increasingly important for drug development in targeted therapies. The use of biomarkers has the potential to facilitate more effective outcomes by guiding patient selection appropriately, thus enhancing benefit-risk profile and improving trial power. Studying a broad population simultaneously with one allows determine which treatment is goal making approved regulatory labeling as inclusive appropriate. We examine new methods accounting complete correlation structure group sequential designs hypotheses nested subgroups. provide full control family-wise Type I error rate. This extension previous either design or between subgroups improves efficiency (power sample size) over typical Bonferroni approach testing populations.

Estimation After a Group Sequential Trial.
Group sequential trials are one important instance of studies for which the sample size is not fixed a priori but rather takes one of a finite set of pre-specified values, dependent on the observed data. Much work has been devoted to the inferential consequences of this design feature. Molenberghs et al (2012) and Milanzi et al (2012) reviewed and extended the existing literature, focusing on a...
متن کاملA Randomized Clinical Trial of Group Cognitive-Rehabilitation Therapy for Patients with Schizophrenia Resistant to Medication
Objectives: Against the background of evidence-based treatments for schizophrenia resistant to medication, the implementation of cognitive-Rehabilitation therapy (CRT) becomes more important, especially about patients who don’t response to medication. There is less evidence for group format and face to face structure of CRT and also for resistant to medication patients. This research investigat...
متن کاملPhase II Basket Group Sequential Clinical Trial with Binary Responses
The basket trial is a recent development in the design of clinical trial. It tests the same treatment on several different related diseases in a single trial and reduces cost and enhances efficiency. The group sequential trial design is commonly used for phase II trials, in which the trial is monitored in several stages and may terminate before the planned end if significant inefficiency is det...
متن کاملAsymptotic multiplicity scaling: a renormalization group perspective
A generalization of the Polyakov-Koba-Nielsen-Olesen scaling law of the multiplicity distributions P (n, s) is developed. It states that a suitable change in the normalization point of P (n, s) compensated by a rescaling can restore data collapsing onto a universal curve if the original scaling rule is violated. We show that the iteratively executed transformation of P (n, s) can be viewed as v...
متن کاملKlotho Protein,A Biomarker for AKI
Klotho is an anti-aging single-pass membrane protein that is mainly produced in the kidney. The level of soluble klotho decreases with age and the klotho gene is associated with an increased risk of age-related diseases, such as diabetes, skin atrophy, chronic kidney disease, ataxia and cancer. The klotho gene is composed of five exons and encodes a membrane glycoprotein located in the plasma ...
